Vitamin D Binding ProteinThink of Transport Proteins as molecular taxis - they collect vitamins, fatty acids & actins then transport them to where they are required. Transport Proteins help the body to rejuvenate naturally by binding and assist with the transportation of, vitamins, fatty acids, and macromolecules found in emu and argan oil and sunlight exposure.   Transport proteins are integral membrane proteins; that is, they exist within and span the membrane across which they transport substances.

Vitamin D Transport Protein

Vitamin D Transport Protein is multi-functional plasma protein with many important functions. The most well known one, and the reason for its name, is the transport of vitamin D metabolites .  Because VDTP is the primary transporter of vitamin D, it has a role in maintaining the total levels of vitamin D in the body and in regulating the amounts of free (unbound) vitamin D available for specific tissues and cell types to utilise.

Other functions of VDTP are to control bone development, binding of fatty acids, sequestration of actin and a range of less-defined roles in modulating immune and inflammatory responses.  The actin removal is considered to be a very important role for VDTP, actins being toxic  and released into the body following cell death.

Two amino acids on this protein chain have been shown to be very strongly associated with the activation of macrophages. In some publications, the vitamin D binding protein is shown to respond to enzymes released by inflammation. Studies show that this isoform not only has the potential to activate macrophages, it can turn them off when no longer needed, so can be classed as an immune system regulator.

Deglosylated Vitamin D TransportProtein (D-VDTP)

We have engineered Vitamin D Transport Protein so that is can deliver it's molecules, vitamins and fatty acids without hindrance from Nagalase.  Nagalase is an enzyme that occurs naturally in the intestines where it breaks down Glycoprotein in our food.

Nagalase is also produced in quantities by cancers, viruses & bacteria to break down a specific glycoprotein in our blood - Vitamin D Transport (Glyco)Protein.  Because it is an enzyme—a catalyst—Nagalase performs this malicious ritual over and over and over again, and each time it comes away unscathed and unchanged. One Nagalase molecule can thus destroy a huge quantity of Vitamin D Binding Protein molecules resulting in immune suppression or collapse.

Nagalase has no natural enemies. No bodily process, no drug, no treatment could outsmart this diabolical killer. Until Dr. Nobuto Yamamoto researched and published Nagalase, we had no idea as to the actual cause of the immune shutdown that allows cancers and viruses grow unchecked.

We use our own enzymes to cleave parts of the Vitamin D Transport Protein which renders it 'immune' against Nagalase therefore it is possible to stimulate the immune system in the presence of chronic illnesses.

Laboratory standards

Cyto Innovations Ltd works in accordance with ISO17025 and GMP European regulations, raw materials are registered and notified to the relevant authorities from where they are purchased.

Lectin activity assay

Biotinylated Vicia Villosa Lectin Assay. Samples are separated in 12% SDS PAGE and analyzed in WB using lectin specific for terminal GalNAc.  Batch should be declared a “PASS” if native blots are observed.

Cell based activity assay

Proliferation assay using RAW264.7 cells. When applied at a level of 40ng/mL to cells during a 72h incubation causes a retardation in cell proliferation when compared to cells that have had no additive or MAF added at the same concentration. The batch should be considered a “PASS” if the cell proliferation is <80% of the D-VDBP treated cells.

 

Sterility testing

Samples of each batch are sent for external sterility testing at a third party testing house. The test is a sterile filtration test which was validated for the procedure to meet the Ph. EU and USP In addition sample material is also incubated in TSB and Thioglycollate Broth between 20-35ºC for 14 days. The batch should be declared a “PASS” if there is complete absence of any bacterial growth in any of the broths or filter test.

Endotoxin detection

Samples of each batch are tested for the presence of endotoxin using a Charles River Endosafe PTS FDA approved endotoxin detection device.  The batch should be declared a “PASS” if there is <0.25 EU/mL. With a 0.25mL dose this is <0.0625EU/dose below the permissible limit of 5EU/Kg body weight.

PH level assessment

PH.Samples of each batch are assessed for pH with limits set between pH7.0-7.9. The batch should be declared a “PASS” if the measured pH is between pH7.0 and pH7.9.
